Meet our Flagship Projects: Discovery of Novel Antimalarial Lead Candidates in Africa

Since its inception in 2023, the Grand Challenges African Drug Discovery Accelerator (GC ADDA) has worked to build a sustainable, Africa-led drug discovery ecosystem by developing and retaining scientific talent, strengthening research capacity, and fostering globally connected centres of excellence across the continent.
While achieving this mission requires many interconnected activities, our four flagship projects sit at the heart of the network. Bringing together researchers from across Africa, these projects are tackling critical scientific challenges while helping to build the skills, infrastructure, and collaborations needed to accelerate drug discovery for unmet medical needs.
To showcase the people and science driving this work, we are launching a new blog series that takes a closer look at each flagship project through an informal Q&A with its leadership team.
In this edition, we find out more about the Discovery of Novel Antimalarial Lead Candidates in Africa flagship project led by Prof. Richard Amewu (University of Ghana, Ghana) and Prof. Lyn-Marie Birkholtz (University of Pretoria and Stellenbosch University, South Africa). The research is conducted in collaboration with GC ADDA Full Members Prof Amanda Rousseau (University of the Witwatersrand, South Africa) and Dr Winston Nxumalo (University of Limpopo, South Africa), as well as GC ADDA Associate Members Dr Laurent Dembele and Dr Dinkorma Ouologuem (University Of Science Of Technical And Technology De Bamako, Mali), Dr Arnold Donkor (Kwame Nkrumah University of Science and Technology, Ghana), Dr Candice Soares De Melo, Dr Liezl Krugmann and Dr Mathew Njoroge (H3D Centre, South Africa), and Dr Mariette van der Watt and Tayla Rabie (University of Pretoria, South Africa).
Share a concise overview of your project and the challenge/s it aims to address.
Our flagship project focuses on discovering new medicines to fight malaria, with a special emphasis on the work of African scientists on the African continent. We aim to develop new drug candidates that can effectively target resistant strains of the malaria parasite and be used as part of global efforts to eliminate the disease. Our team searches for new chemical compounds—small molecules—that can kill malaria parasites grown in the laboratory and in field isolates. We then work to improve these compounds on the path to optimised potential medicines, ensuring they are safe for humans and have a high likelihood of being effective in future treatments.
Why is this kind of research crucial, particularly in an African context?
Africa still bears the biggest share of the world's malaria problems, and the most at risk are children under 5 and pregnant women. It’s crucial that we continue to find ways to save their lives. Sadly, the medicines we rely on in clinics and hospitals to prevent death from malaria are facing growing resistance, making them less effective. We need to develop new medicines quickly, especially those tailored for African communities. Luckily, Africa has the scientific talent to lead this effort. Our discoveries can help create medicines made right here at home, which will be readily available to those in need.
How has the project contributed to strengthening the capacity for drug discovery research within Africa so far?
Our project follows examples of classical drug discovery pipelines that we have expanded and consolidated into a coherent project, spread across partners across the continent. The investment made in the project by the Gates Foundation and LifeArc has enabled the expansion of existing infrastructure in Ghana and strengthened human capacity, with around 8 expert chemists and 5 expert biologists trained and mentored in advanced, industry-level drug discovery in Ghana and South Africa. These emerging scientists also had the opportunity to attend Drug Discovery workshops in Ghana and South Africa and to present their work at international conferences. More than 3 researchers underwent international training at the University of Dundee in Scotland and at the Medicines for Malaria Venture in Switzerland, bringing back technologies and expertise to enrich our current efforts on the continent.
What are the benefits of working within a network such as GC ADDA, particularly for advancing drug discovery on the continent?
The GC ADDA network has been extraordinarily important to the success of our project. The network enabled like-minded researchers to come together to formulate the research project in a way that supported competitive, industry-standard drug discovery. The partners that the GC ADDA network brings to the table have expanded our opportunities and capabilities and provided access to bespoke, high-quality workshops. Moreover, the network has provided access to licenced data analysis platforms and unique development platforms.
What would you like to see from African governments and institutions to better support drug discovery and innovation on the continent?
We have to support sustainability in early drug discovery with in-house investments. Africans will make up 1 in 4 people by 2050, so we have to invest in protecting this human capital. African governments can support GC ADDA by co-investing in hubs as centres of excellence to strengthen the continent's drug discovery portfolio.
