      <title>Meet Our Flagship Projects: African Drug Discovery Natural Products Box</title>

           Since its inception in 2023, the Grand Challenges African Drug Discovery Accelerator (GC ADDA) has worked to build a sustainable, Africa-led drug discovery ecosystem by developing and retaining scientific talent, strengthening research capacity, and fostering globally connected centres of excellence across the continent.
          &#xD;
    &lt;/span&gt;&#xD;
  &lt;/p&gt;&#xD;
  &lt;p&gt;&#xD;
    &lt;span&gt;&#xD;
      &lt;br/&gt;&#xD;
    &lt;/span&gt;&#xD;
  &lt;/p&gt;&#xD;
  &lt;p&gt;&#xD;
    &lt;span&gt;&#xD;
      
           While achieving this mission requires many interconnected activities, our four flagship projects sit at the heart of the network. Bringing together researchers from across Africa, these projects are tackling critical scientific challenges while helping to build the skills, infrastructure, and collaborations needed to accelerate drug discovery for unmet medical needs.
          &#xD;
    &lt;/span&gt;&#xD;
  &lt;/p&gt;&#xD;
  &lt;p&gt;&#xD;
    &lt;span&gt;&#xD;
      &lt;br/&gt;&#xD;
    &lt;/span&gt;&#xD;
  &lt;/p&gt;&#xD;
  &lt;p&gt;&#xD;
    &lt;span&gt;&#xD;
      
           To showcase the people and science driving this work, we are launching a new blog series that takes a closer look at each flagship project through an informal Q&amp;amp;A with its leadership team.
          &#xD;
    &lt;/span&gt;&#xD;
  &lt;/p&gt;&#xD;
  &lt;p&gt;&#xD;
    &lt;span&gt;&#xD;
      &lt;br/&gt;&#xD;
    &lt;/span&gt;&#xD;
  &lt;/p&gt;&#xD;
  &lt;p&gt;&#xD;
    &lt;span&gt;&#xD;
      
           We begin with the African-derived Natural Products Box (AdNPB), based at the University of Buea in Cameroon and led by Principal Investigator Prof Fidele Ntie-Kang.
          &#xD;
    &lt;/span&gt;&#xD;
  &lt;/p&gt;&#xD;
  &lt;p&gt;&#xD;
    &lt;span&gt;&#xD;
      &lt;br/&gt;&#xD;
    &lt;/span&gt;&#xD;
  &lt;/p&gt;&#xD;
  &lt;p&gt;&#xD;
    &lt;strong&gt;&#xD;
      
           Share a concise overview of your project and the challenge/s it aims to address. 
          &#xD;
    &lt;/strong&gt;&#xD;
  &lt;/p&gt;&#xD;
  &lt;p&gt;&#xD;
    &lt;span&gt;&#xD;
      
            
          &#xD;
    &lt;/span&gt;&#xD;
  &lt;/p&gt;&#xD;
  &lt;p&gt;&#xD;
    &lt;span&gt;&#xD;
      
           The AdNPB (African Drug Discovery Natural Products Box) Flagship Project is creating a carefully selected collection of natural compounds obtained from African plants and other natural sources to support the discovery of new medicines. Scientists reviewed and validated 300 compounds, assessing their chemical diversity and suitability for further development. From these, 100 high-quality compounds were selected for inclusion in the AdNPB collection. 
          &#xD;
    &lt;/span&gt;&#xD;
  &lt;/p&gt;&#xD;
  &lt;p&gt;&#xD;
    &lt;span&gt;&#xD;
      &lt;br/&gt;&#xD;
    &lt;/span&gt;&#xD;
  &lt;/p&gt;&#xD;
  &lt;p&gt;&#xD;
    &lt;span&gt;&#xD;
      
           One of the major challenges facing drug discovery research in Africa is limited access to advanced scientific equipment needed to fully identify and evaluate promising compounds. To address this, the project works closely with partner laboratories that have specialized facilities. These collaborations allow African researchers to access advanced technologies, strengthen scientific partnerships, and build local research capacity while contributing to global drug discovery efforts. 
          &#xD;
    &lt;/span&gt;&#xD;
  &lt;/p&gt;&#xD;
  &lt;p&gt;&#xD;
    &lt;span&gt;&#xD;
      &lt;br/&gt;&#xD;
    &lt;/span&gt;&#xD;

           Why is this kind of research crucial, particularly in an African context? 
          &#xD;
    &lt;/strong&gt;&#xD;

           &#xD;
      &lt;br/&gt;&#xD;

           Africa is home to an extraordinary diversity of plants and other natural resources that could provide the foundation for future medicines. However, much of this potential remains underexplored. 
          &#xD;
    &lt;/span&gt;&#xD;
  &lt;/p&gt;&#xD;
  &lt;p&gt;&#xD;
    &lt;span&gt;&#xD;
      &lt;br/&gt;&#xD;
    &lt;/span&gt;&#xD;
  &lt;/p&gt;&#xD;
  &lt;p&gt;&#xD;
    &lt;span&gt;&#xD;
      
           Research projects such as the AdNPB project helps African scientists identify, study, and test promising natural substances locally rather than relying entirely on discoveries made elsewhere. It also helps develop scientific expertise, strengthens research institutions, and creates opportunities for African researchers to contribute directly to solutions for diseases that affect populations on the continent and beyond. 
          &#xD;
    &lt;/span&gt;&#xD;
  &lt;/p&gt;&#xD;
  &lt;p&gt;&#xD;

           By investing in the study of Africa’s natural resources, we can transform biodiversity into knowledge, innovation, and potentially life-saving medicines.

           How has the project contributed to strengthening the capacity for drug discovery research within Africa so far?

           The project is helping to build drug discovery capacity by training researchers in the isolation, characterisation, and biological evaluation of purified natural compounds from plants and other sources. 
          &#xD;
    &lt;/span&gt;&#xD;
  &lt;/p&gt;&#xD;
  &lt;p&gt;&#xD;
    &lt;span&gt;&#xD;
      &lt;br/&gt;&#xD;
    &lt;/span&gt;&#xD;

    &lt;span&gt;&#xD;
      
           It has established a curated collection of well-characterised compounds, implemented quality-control procedures using available laboratory tools, and created pathways for sharing samples with partner laboratories for advanced biological screening. Through these activities, researchers gain valuable practical experience while strengthening networks that support collaborative science across Africa and internationally.

           The Grand Challenges African Drug Discovery Accelerator (GC ADDA) provides an important platform for collaboration, training, and knowledge sharing among African scientists.

           Through GC ADDA, researchers gain access to shared digital tools that improve research management and data handling. Workshops, internships, mentorship programmes, and collaborative projects help strengthen skills and foster lasting scientific partnerships across the continent.

           For example, members of our team have benefited from the H3D Global Health Mentorship Programme, gaining valuable experience and exposure to international best practices in drug discovery research.

  &lt;p&gt;&#xD;
    &lt;span&gt;&#xD;
      &lt;br/&gt;&#xD;
    &lt;/span&gt;&#xD;

           What would you like to see from African governments and institutions to better support drug discovery and innovation on the continent? 
          &#xD;

    &lt;span&gt;&#xD;
      
            
          &#xD;
    &lt;/span&gt;&#xD;

           To accelerate drug discovery and innovation, African governments and institutions should invest in modern research infrastructure and provide sustained long-term funding for scientific research. 
          &#xD;
    &lt;/span&gt;&#xD;
  &lt;/p&gt;&#xD;
  &lt;p&gt;&#xD;
    &lt;span&gt;&#xD;
      &lt;br/&gt;&#xD;
    &lt;/span&gt;&#xD;
  &lt;/p&gt;&#xD;
  &lt;p&gt;&#xD;
    &lt;span&gt;&#xD;
      
           There is also a need to train more scientists through hands-on programmes that develop practical drug discovery skills. Improving research data management systems, promoting collaboration among African institutions, and strengthening partnerships with global research organizations would further enhance scientific productivity. 
          &#xD;
    &lt;/span&gt;&#xD;
  &lt;/p&gt;&#xD;
  &lt;p&gt;&#xD;
    &lt;span&gt;&#xD;
      &lt;br/&gt;&#xD;
    &lt;/span&gt;&#xD;
  &lt;/p&gt;&#xD;
  &lt;p&gt;&#xD;
    &lt;span&gt;&#xD;
      
           Importantly, universities should introduce integrated drug discovery training programmes at Master's and PhD levels to prepare the next generation of researchers with the multidisciplinary skills needed to transform scientific discoveries into health solutions that benefit African populations.

            Earlier this year, the South African Medical Research Council (SAMRC) committed R6 million in funding to support two early-stage drug discovery research platforms under the GC ADDA banner.
           &#xD;
      &lt;/span&gt;&#xD;
    &lt;/span&gt;&#xD;
  &lt;/p&gt;&#xD;
  &lt;p&gt;&#xD;
    &lt;span&gt;&#xD;
      &lt;br/&gt;&#xD;
    &lt;/span&gt;&#xD;
  &lt;/p&gt;&#xD;
  &lt;p&gt;&#xD;
    &lt;span&gt;&#xD;
      
           Both projects are hosted by North-West University (NWU) in Potchefstroom, South Africa and are aimed at strengthening early-stage Gram-negative antibiotic discovery and toxicology capability within the GC ADDA research pipeline. The grants were awarded to projects led by Dr Fortunate Mokoena and Dr Rose Hayeshi, both emerging African women principal investigators and GC ADDA Full Members, underscoring a commitment to advancing women’s leadership in science, strengthening early-career research capacity, and promoting inclusive participation in biomedical research
          &#xD;
    &lt;/span&gt;&#xD;
  &lt;/p&gt;&#xD;
  &lt;p&gt;&#xD;
    &lt;span&gt;&#xD;

           Identification and Characterization of Hits Against Carbapenem-Resistant Klebsiella pneumoniae

    &lt;/span&gt;&#xD;

           is led by Dr Fortunate Mokoena, senior lecturer in NWU's Department of Biochemistry.

           Can you provide a brief overview of your GC ADDA, SAMRC-funded project and the problem it aims to address?
          &#xD;
    &lt;/strong&gt;&#xD;
  &lt;/p&gt;&#xD;
  &lt;p&gt;&#xD;
    &lt;span&gt;&#xD;
      &lt;br/&gt;&#xD;
    &lt;/span&gt;&#xD;
  &lt;/p&gt;&#xD;
  &lt;p&gt;&#xD;
    &lt;strong&gt;&#xD;
      
           FM:
          &#xD;
    &lt;/strong&gt;&#xD;
    &lt;span&gt;&#xD;
      
           This project addresses the growing global threat of antimicrobial resistance (AMR), where disease-causing microorganisms are becoming increasingly able to resist treatment with currently available antibiotics. As a result, infections are becoming more difficult to treat, highlighting the urgent need for new and effective antimicrobial therapies. We propose a drug repurposing approach that utilises FDA-approved drugs, which have already undergone extensive safety and efficacy testing. This strategy aims to accelerate the development process and improve the likelihood of successful treatment options for resistant infections.

    &lt;/span&gt;&#xD;
  &lt;/p&gt;&#xD;
  &lt;p&gt;&#xD;
    &lt;br/&gt;&#xD;
  &lt;/p&gt;&#xD;
  &lt;p&gt;&#xD;

           The project aims to build toxicology capability to support GC ADDA Flagship projects. The project will address a critical gap in toxicology capability and expertise within GC ADDA through targeted training, expert advisory support, and structured knowledge exchange. The project is designed to enable the practical integration of early investigational toxicology assessments into ongoing GC ADDA projects while laying the foundation for sustained toxicology capability across participating African institutions.

    &lt;span&gt;&#xD;

           Why is this kind of work so crucial in the fight against AMR, particularly in the African context?
          &#xD;
    &lt;/strong&gt;&#xD;
  &lt;/p&gt;&#xD;
  &lt;p&gt;&#xD;
    &lt;span&gt;&#xD;
      &lt;br/&gt;&#xD;
    &lt;/span&gt;&#xD;
  &lt;/p&gt;&#xD;
  &lt;p&gt;&#xD;
    &lt;strong&gt;&#xD;
      
           FM:
          &#xD;
    &lt;/strong&gt;&#xD;
    &lt;span&gt;&#xD;
      &lt;span&gt;&#xD;
      &lt;/span&gt;&#xD;

    &lt;span&gt;&#xD;
      
           In 2019, the World Health Organization reported that AMR caused approximately 1.27 million deaths globally, with Sub-Saharan Africa being the most affected region. South Africa specifically recorded about 35,000 deaths due to AMR in 2021. The rampant use of antibiotics, particularly in intensive care units across both public and private healthcare sectors, has intensified the problem of drug resistance. Notable pathogens contributing to this crisis include Klebsiella pneumoniae, Acinetobacter baumannii, and Candida auris. The effectiveness of carbapenems, often reserved as a last resort for treating severe bacterial infections, is diminishing. To address this urgent issue, our research focuses on developing new therapeutic strategies targeting carbapenem-resistant Klebsiella pneumoniae, a highly prevalent and significant clinical threat in South Africa. 
          &#xD;
    &lt;/span&gt;&#xD;
  &lt;/p&gt;&#xD;
  &lt;p&gt;&#xD;
    &lt;span&gt;&#xD;
      &lt;br/&gt;&#xD;
    &lt;/span&gt;&#xD;
  &lt;/p&gt;&#xD;
  &lt;p&gt;&#xD;
    &lt;strong&gt;&#xD;

           Toxicology, the study of potential harm caused by substances, is essential to evaluating whether candidate drugs are safe enough to proceed to clinical trials and is a major cause of failure of new drug molecules making it to the market. Toxicology is a critical component of drug discovery, enabling early risk identification, compound triage, and informed project decision‑making. Therefore, this work is critical in the fight against AMR, as it will directly influence the progression and quality of candidate compounds in the development of drugs to combat AMR.
          &#xD;
    &lt;/span&gt;&#xD;
  &lt;/p&gt;&#xD;
  &lt;p&gt;&#xD;
    &lt;span&gt;&#xD;
      &lt;br/&gt;&#xD;
    &lt;/span&gt;&#xD;
  &lt;/p&gt;&#xD;
  &lt;p&gt;&#xD;
    &lt;span&gt;&#xD;
      &lt;br/&gt;&#xD;
    &lt;/span&gt;&#xD;

           What kind of capacity-strengthening opportunities will come out of your project? And what does this mean for building sustainable drug discovery capacity in Africa?

           This funding significantly strengthens capacity development in Africa by supporting the training of a PhD candidate, an MSc student, and a post-doctoral fellow who will also help mentor junior scientists. It enhances collaboration with the University of Cape Town and H3D, where Co-PI Dr Drik Lamprecht will provide advanced training in key areas such as broad-spectrum activity profiling and CRISPR-Cas target validation.

  &lt;/p&gt;&#xD;
  &lt;p&gt;&#xD;
    &lt;span&gt;&#xD;

           We hope that this project will also benefit from engagement with GC ADDA training opportunities, helping to build a stronger network and a critical mass of skilled drug discovery scientists across Africa.

           The project will train 1-2 trainees, the main one being an emerging researcher with post-PhD experience. GC ADDA team members from the DMPK flagship project will also have the opportunity to take part in training. As Africa moves towards securing its ability to develop drugs from bench to bedside, it is vital to strengthen the skills in toxicology that will be necessary to support this critical endeavour.
